No. Princess Peach is not Bowser Jr.’s mother. In Super Mario Sunshine, Bowser tells his son that Peach is his mother, but the ending makes clear that this was a story Bowser had told him. Bowser Jr.’s actual mother is not identified.
Why does Bowser Jr. call Peach “Mama Peach”?
In Super Mario Sunshine, Bowser Jr. appears as Shadow Mario and acts on what Bowser has told him: that Peach is his mother and that Mario kidnapped her. That claim explains Jr.’s behavior in the story; it is not evidence that Peach is his parent. The game’s dialogue is transcribed by Super Mario Wiki.
What does the ending reveal?
Near the end, Bowser begins a difficult explanation about Princess Peach. Bowser Jr. responds, “I know she’s not really my Mama…” The exchange confirms that Peach is not really his mother, while leaving the identity of his actual mother unanswered.
Who is Bowser Jr.’s mother?
The available answer from creator commentary is that her identity is unknown. In an interview republished by Game Informer on September 10, 2015, Shigeru Miyamoto said, “Bowser’s only child is Bowser Jr., and we do not know who the mother is.” The comment does not name a character or offer a hidden explanation.

Did Miyamoto say he was Bowser Jr.’s mother?
A 2015 Nintendo UK myth-busting video prompted a joking response that Miyamoto was Bowser Jr.’s mother. GameSpot’s September 10, 2015 coverage presented the remark as a gag. It should not be read as a literal reveal or as a resolution of the character’s parentage.
