Guillermo does become a vampire in Season 5
The question is really about why Guillermo is not still a vampire. FX’s Season 5 episode listing, “Guillermo (Finally) Becomes a Vampire,” describes him transforming and gaining power and immortality. His long wait for the change is fulfilled—but the reality of vampire life proves different from the fantasy.

Why does Guillermo give up being a vampire?
The clearest explanation is personal: Guillermo cannot accept being a bloodthirsty predator. FX’s cast description says that after he is turned, he “discovered he didn’t have what it took to be a bloodthirsty monster.” Becoming a vampire gives him the supernatural status he once desired, but it also requires a way of living that he cannot stomach.
Does Van Helsing ancestry prevent him from becoming a vampire?
No confirmed rule in the show’s official descriptions says Guillermo’s Van Helsing heritage makes transformation impossible. His ancestry is discussed in a TV Insider interview about the Season 5 finale as a possible complication, but it cannot be the definitive explanation for why he is not a vampire: FX confirms that he does transform. The more clearly established reason he does not remain one is that he cannot embrace vampire bloodlust.

Is Guillermo human by the end of the series?
Yes. ABC’s series overview describes him as the vampires’ “former familiar and current human friend.” That later description makes the distinction clear: he was briefly transformed, but his eventual place in the group is as a human.
