Spider-Man’s best-known line is the responsibility motto: “With great power comes great responsibility.” Its original comic wording is slightly longer, and it appears in narration—not as dialogue spoken by Uncle Ben in that 1962 story. A second familiar phrase is “Your friendly neighborhood Spider-Man.”

“With great power” is Spider-Man’s defining motto

The familiar short form, “With Great Power Comes Great Responsibility!”, appears as Peter Parker’s associated wisdom on Marvel’s official Spider-Man character page. It is the clearest answer to the question of Spider-Man’s most famous quote, though no survey or ranking establishes it as an objectively measured number one.

The original comic uses a longer wording

Spider-Man debuted in Amazing Fantasy #15 in August 1962, as documented by the Library of Congress. The story’s closing narration reads: “With great power there must also come great responsibility.” That longer version is part of the concluding narration; it is not a line spoken by Uncle Ben in the original story.

Why both versions are quoted

The two wordings express the same principle, but they are not identical quotations. The comic’s original narration includes “there must also,” while Marvel’s character page uses the shorter form now commonly associated with Spider-Man. When accuracy to the 1962 story matters, use the longer wording and identify it as narration; when referring to the familiar motto, the concise form is appropriate.

“Your friendly neighborhood Spider-Man”

“Your friendly neighborhood Spider-Man” is another recognizable catchphrase. A Wiley excerpt from Origin Stories prints the phrase among superhero catchphrases. That source supports its use as a catchphrase, but does not identify a first appearance, origin scene, or particular speaker. It is best presented as a familiar Spider-Man phrase rather than assigned to a specific comic, film, or scene without further evidence.

What the sources establish—and what they don’t

The Library of Congress documents the original comic’s date and concluding wording, while Marvel’s character page supplies the familiar concise motto. Wiley provides evidence for the neighborhood catchphrase. Together, these sources support a curated selection of recognizable lines, not a complete ranking across comics, films, television, and games. They also do not justify assigning either motto to a particular screen adaptation or scene beyond the contexts described above.
