Usually, not in the way creators mean. Amazon’s documented self-service route for selling digital books is Kindle Direct Publishing (KDP), which distributes ebooks through Kindle Stores. The answer depends on what “without Kindle” means: you can publish through KDP without owning a Kindle device, but the reviewed Amazon sources do not establish a general self-service option for selling arbitrary downloadable files—such as PDFs, templates, courses, or software—directly on Amazon without using its Kindle publishing and store system.
What “without Kindle” can mean
There are two different questions hidden in the phrase. If you mean you do not own a Kindle device, that does not rule out publishing an ebook through KDP: Amazon’s publishing guidance refers to reading on Kindle devices and apps. If you mean you want to avoid listing the ebook in the Kindle Store altogether, KDP is not a Kindle-free route. Amazon describes KDP as its self-service publishing option for ebooks and print books, with ebook distribution through Kindle Stores (KDP; KDP publishing overview).

If you want to sell an ebook on Amazon and elsewhere
Ordinary KDP publishing and KDP Select are not the same thing. Amazon’s help identifies KDP Select enrollment as the condition that makes the ebook exclusive to Amazon. Do not assume that exclusivity applies to every KDP ebook; check the current program terms before enrolling or distributing the same ebook through other retailers (KDP Select help).
If you want to sell a PDF, template, course, or software download
The reviewed official material does not establish that ordinary Seller Central offers general direct delivery of arbitrary downloadable products. A Seller Central community discussion reports restrictions on standalone digital goods and downloadable software, but a forum discussion is not the governing policy and does not settle possible exceptions or eligible categories (Seller Central community discussion). Do not treat a physical product listing or an ebook listing as a workaround for delivering a separate file. Check the current official policy for the specific product and marketplace before attempting to list it.

If you have audiobook rights
ACX may be relevant if your ebook is already published in the Kindle Store and you hold the necessary audiobook rights. Its help page describes that Kindle ebook prerequisite; ACX is therefore an adjacent audiobook option, not a way to avoid Kindle publishing (ACX eligibility requirements; ACX producer information).

Amazon’s physical-book routes are a different business model: the customer buys and receives a physical book, not the downloadable original file. Amazon’s guidance distinguishes KDP publishing from Seller Central and Amazon Advantage for physical book inventory (Amazon seller account information). A physical listing does not create Amazon-hosted delivery for a PDF, design file, course, or software package.
