There is no verified public explanation for why Princess Daisy was not included in the 2023 animated The Super Mario Bros. Movie. Nintendo and Illumination’s announcement names the principal cast without Daisy, but it does not say why she was left out.
What the official cast announcement tells us
Nintendo and Illumination described the film as “the new animated film based on the world of Super Mario Bros.” In their March 10, 2023 announcement, they named Chris Pratt as Mario, Anya Taylor-Joy as Princess Peach, Charlie Day as Luigi, and other principal cast members, but not Daisy. Illumination’s film page also does not name her.

Those materials establish that Daisy was not among the announced principal cast. They do not document the reason for that choice, and the reviewed film materials offer no explanation from the filmmakers. Daisy and Peach are different characters
Daisy is not another name for Peach. They are separate characters in the Mario series, and Daisy’s roots are associated with the Super Mario Land side-series, while Peach belongs to the core series. That distinction may help explain why viewers notice Daisy’s absence, but it does not establish why the film’s creative team chose its cast.

Daisy also appeared in the 1993 live-action Super Mario Bros. film. That earlier movie is distinct from the 2023 animated film, so memories of Daisy in a “Mario movie” can refer to a different production. (Character background and film history)
What about the later Mario movie?
Later coverage reports that Daisy appears in a second post-credits scene of The Super Mario Galaxy Movie. That is a separate, later film and does not change what was announced for the 2023 movie.
